ABSTRACT

OBJECTIVE@#To analyze the clinical phenotype and genetic etiology for a child featuring congenital hypothyroidism (CH).@*METHODS@#Whole exome sequencing (WES), copy number variation (CNV) sequencing and chromosomal microarray analysis (CMA) were carried out for a newborn infant who had presented at Linyi People's Hospital for CH. Clinical data of the child was analyzed, in addition with a literature review.@*RESULTS@#The main characteristics of the newborn infant had included peculiar face, vulvar edema, hypotonia, psychomotor retardation, recurrent respiratory tract infection with laryngeal wheezing and feeding difficulties. Laboratory test indicated hypothyroidism. WES suggested a CNV deletion on chromosome 14q12q13. CMA further confirmed a 4.12 Mb deletion at chromosome 14q12q13.3 (32649595_36769800), which has encompassed 22 genes including NKX2-1, the pathogenic gene for CH. The same deletion was found in neither of her parents.@*CONCLUSION@#Through the analysis of clinical phenotype and genetic variant, the child was diagnosed with 14q12q13.3 microdeletion syndrome.

ABSTRACT

OBJECTIVE To analyze the dose-adjusted concentrations of Posaconazole oral suspension in patients undergoing hematopoietic stem cell transplantation (HSCT) and their influential factors. METHODS Data were collected from hospitalized HSCT patients admitted to the First Affiliated Hospital of Shandong First Medical University (Shandong Provincial Qianfoshan Hospital) from January 2021 to April whtwhm@yeah.net 2023 who took Posaconazole oral suspension for the prevention of invasive fungal disease (IFD) and received blood concentration of posaconazole. The rate of concentration attainment and clinical failure rate of posaconazole for the prevention of IFD were evaluated, and one-way and multiple linear regression analyses were performed for the influential factors of dose-adjusted concentrations (C0/D) of posaconazole. RESULTS A total of 44 patients were enrolled; the mean C0 of posaconazole in patients was (0.99±0.94) µg/mL, and 20 patients had a C0≥0.7 μg/mL, with a concentration attainment rate of 45.45% for the prevention of IFD; 13 cases were clinical failures, with a clinical failure rate of 29.55%. Of 24 patients who did not achieve C0/D of posaconazole for IFD prophylaxis, one patient was a clinical failure despite timely dose adjustment of posaconazole in seven patients; seven of the thirteen patients who did not undergo dose adjustment were clinical failures; and the remaining four patients were switched to other antifungal agents. The results of univariate analysis showed that gender, body mass index (BMI), renal function, combined use of sodium phenytoin, omeprazole and metoclopramide had a significant effect on the C0/D of posaconazole (P<0.05); the results of multivariate linear regression analysis showed that gender, BMI and combined use of sodium phenytoin were the independent factors affecting the C0/D of posaconazole (P<0.05). CONCLUSIONS Significant individual differences are reflected in the blood concentration of Posaconazole oral suspension; gender, BMI and combined use of sodium phenytoin are independent factors affecting the C0/D of posaconazole.

ABSTRACT

Objective To observe the expression difference of lncRNA FAL1 in ovarian cancer cells and their drug-resistant cell lines, and to explore the effect and mechanism of lncRNA FAL1 down-regulation on cell chemotherapy resistance. Methods The expression levels of fal1 gene in SKOV3 and COC1 cells and their drug-resistant cell lines were detected by qRT-PCR. fal1 siRNA was transfected to downregulate fal1 gene expression. MTT was used to detect cell proliferation. Transwell method was used to detect cell invasion ability. Plate clone formation test was used to detect cell clone ability, and Western blot was used to detect MDR-1, mpr-1, ABCG2 and phosphorylation levels of p38 MAPK, ERK1/2 and JNK. SKOV3/DDP and COC1/DDP cells transfected with FAL1-siRNA were injected subcutaneously into BALB/c nude mice. The volume and mass of subcutaneous transplanted tumors were measured. Results Compared with SKOV3 and COC1 cells, SKOV3/DDP and COC1/DDP cells were less sensitive to DDP, and the expression levels of FAL1 gene increased (P < 0.01). After transfection with FAL1-siRNA, the sensitivity of SKOV3/DDP and COC1/DDP cells to DDP increased (P < 0.01), and the invasion (P < 0.05) and cloning ability (P < 0.01) decreased. The expression levels of MDR-1, MPR-1, ABCG2 (P < 0.01) and the phosphorylation levels of p38 MAPK, ERK1/2 and JNK (P < 0.05) decreased. The volume and mass of subcutaneous transplanted tumors were significantly reduced (P < 0.01). Conclusion Down-regulation of lncRNA FAL1 could significantly reduce the chemotherapy resistance of cisplatin-resistant ovarian cancer cell lines and inhibit the proliferation of drug-resistant cells in vivo. Its mechanism is related to inhibiting the activation of MAPK signaling pathway.

Objective@#To explore the effect of quality control circle(QCC)on reducing the incidence of moderate or severe malnutrition in gastric cancer patients under chemotherapy.@*Methods@#Before the activity (February to March 2016), 146 gastric cancer patients under chemotherapy were selected as the control group, and regular nutrition nursing (dietary guidance and regular nutrition-related health lectures) was given. After the activity (February to March 2017), 139 gastric cancer patients under chemotherapy were selected as QCC group, and the standard intervention of quality control circle was implemented. The nutritional status of patients before and after improvement was compared.@*Results@#After finding the cause and formulating and implementing the measures, the 24-hour consultation rate, the incomplete remission rate of nausea/vomiting, the incidence of cancer anorexia, and the rate of missed visits in QCC group were significantly different with those in control group (P<0.05). The incidence of moderate or severe malnutrition was 16.55% in QCC group and 65.07% in control group, and the difference was statistically significant (P<0.01).@*Conclusions@#The quality control circle activity can effectively reduce the incidence of moderate or severe malnutrition and improve the quality of life in gastric cancer patients under chemotherapy. It can also improve the self-discipline and initiative of medical staff and improve the quality of medical care.

ABSTRACT

Objective :To explore influence of cardiopulmonary endurance improvement on blood lipid ,aiming at pro‐viding reference for reducing morbidity of cardiovascular Dis in middle‐aged and aged people in China .Methods :Databases including China National Knowledge Infrastructure (CNKI ) , Wanfang , VIP , Bailianyun etc .were searched .Literature were screened according to inclusion and exclusion criteria ,while article quality assessment and data extraction were performed ,and RevMan5. 3 software was used to perform Meta‐analysis.Results :Meta‐analy‐sis indicated that exercise‐improving cardiopulmonary endurance in middle‐aged and aged people can significantly re‐duce levels of triglyceride [TG ,combined effect size :-0. 42 ,95% CI (-0.60 ,-0.24 ) , P= 0. 001 ] and low density lipoprotein cholesterol [(LDL‐C ,combined effect size :-0.44 ,95% CI (-0.85 , 0. 03) ,P=0.04] ,while it had no significant influence on levels of total cholesterol [TC ,combined effect size :-0.71 , 95% CI (-1. 46 , 0.03) , P=0. 06] and high density lipoprotein cholesterol [HDL‐C ,combined effect size :0. 20 , 95% CI (-0. 09 , 0.50) , P=0.18].Subgroup analysis indicated that power bicycle and type K 4b2 cardiopulmonary endurance meas‐urement method of COSME company (K4b2 method) possessed significant influence on levels of TC and TG ;Bruce treadmill testing system possessed significant influence on HDL‐C level ;Bruce treadmill test and K4b2 method pos‐sessed significant influence on LDL‐C level .Conclusion :Exercise‐improving cardiopulmonary endurance in middle‐aged and aged people can effectively reduce levels of triglyceride and low density lipoprotein cholesterol .

ABSTRACT

Objective To assess the effect of aerobic exercise on the cardiovascular autonomic regulation in healthy elderly people living in hypoxia areas at high altitude,through observing the 24 hours of heart rate variability(HRV) and its changes in HRV before,during and after incremental exercise.Methods 60 healthy elderly(> 60 years old)living in Qinghai-Tibet plateau(altitude > 2 600 m)were selected and divided into aerobic exercise group(practicing Tibetan traditional dance in long term,n=30)and control group (lacking aerobic exercise,n =30).Blood pressure,heart rate (HR) and oxygen saturation (SO2) were measured.The time domain parameters of HRV were detected by 24 hour Holter test.The time domains of HRV before,during and after incremental exercise were observed by Marquette 2000 treadmill exercise test.Results Systolic blood pressure,diastolic blood pressure and heart rate were lower(t=2.475,2.295,2.099,all P<0.05),and SO2 and SO2/HR ratio was higher(t=2.907,2.210,P<0.05)in aerobic exercise group than in control group.As compared with control group,the aerobic exercise group showed that the changes in SDNN were increased significantly during and 10 minutes after incremental exercise(both P<0.05),and RMSSD and PNN50 were significantly decreased during and 10 minutes after exercise(both P<0.05).Conclusions Regular aerobic exercise in the elderly people living in hypoxia areas at high altitude can enhance oxygen uptake and utilization,increase the heart rate variability,reduce heart rate and blood pressure,and improve the cardiac autonomic function.

ABSTRACT

Objective To observe the effect of rehabilitation on ADL scores in patients with type 2 diabetes complicated with cerebral stroke.Methods According to the selected order,three hundred and twenty type 2 diabetes patients complicated with cerebral stroke were numbered in Arabic numbers 1,2,3 and so on,the odd number patients were randomly enrolled into the observation group,the even number patients were assigned into the control group.The control group was only given corresponding clinical treatments.In addition to the same clinical treatments,rehabilitation training was given to the observation group at the same time.The ADL scores,change between the two groups were compared after three months.Results After treatment,the ADL score of the observation group was significantly improved,the difference was statistically significant (t =-24.9,P =0.000,P < 0.05).The ADL score of the control group was also improved,and the difference was statistically significant (t =-15.52,P =0.000,P < 0.05).The ADL score between the observation group and the control group had statistically significant difference (t =24.24,P =0.000,P < 0.05).Conclusion Type 2 diabetes patients complicated with cerebral stroke who are early given rehabilitation training actively,car significantly improve the function of hemiplegia limbs,improve the ADL scores and the quality of life.

ABSTRACT

Objective To observe the clinical effect of psychological intervention combined with alendronate and low molecular heparin calcium on the treatment of senile osteoporotic fracture. Methods From February 2014 to March 2017, clinical data of 82 elderly patients with osteoporotic fracture in the first people's hospital in Jiande were retrospectively analyzed. All the patients were randomly divided into the control group and the experimental group with 41 cases in each group. The control group patients were given alendronate sodium and low molecular heparin calcium; At this basis, the experimental group were received psychological nursing intervention. The effect in the two groups was compared. Results After treatment, bone mineral density (BMD) index and pain scores in the two groups were significantly improved. BMD and pain scores in the experimental group were significantly better than those in the control group (P<0.05). The total effective rate was 95.12% (39/41) and the complication rate was 17.07% (7/41) in the experimental group, while in the control group were respectively 70.73% (29/41), 36.59% (15/41), the differences were statistically significant (P<0.05). Conclusion Psychological intervention combined with alendronate sodium and low molecular heparin on the treatment of senile osteoporotic fracture has significant effect, can effectively improve the clinical symptoms of patients, reduce the incidence of complications, with clinical application value.

ABSTRACT

Objective To assess the expression and significance of proprotein convertase subtilisin kexin 9 (PCSK9) in patients with rheumatoid arthritis (RA).Methods Sixty-five RA patients and forty-seven healthy controls were recruited in this study.The body mass index (BMI) and serum total cholesterol (TC),triglyceride (TG),high density lipoprotein (HDL),lipoprotein a,low density lipoprotein (LDL),very low density lipoprotein(VLDL),apolipoprotein A (ApoA),apolipoprotein B (ApoB) and the ratio of LDL-C/HDL-C were tested.Other parameters included disease activity score 28 (DAS28),rheumatoid factor (RF),anti-cyclic citrullinated peptide (CCP) antibody,erythrocyte sedimentation rate (ESR),c-reactive protein (CRP).Serum PCSK9 level was measured by ELISA and compared between RA patients and healthy controls.Results (1) The serum PCSK9 levels in RA patients were higher than those in healthy controls [(409.36 ±223.52) μg/L vs (292.19 ± 109.79) μg/L,P < 0.05].(2) Compared with subgroup of moderate and low active disease and patients in remission,PCSK9 was significantly higher in patients with highly active disease (P < 0.05).(3) The serum PCSK9 levels were positively correlated with RF,TC,TG,LDL,very low density lipoprotein (VLDL),ApoB,with r values as 0.303,0.490,0.320,0.451,0.319,0.463,respectively (P < 0.05).(4) Multiple stepwise regression analysis showed that DAS28,RF,TC and LDL-C/HDL-C were relevant factors for PCSK9 in RA patients.Conclusions The serum PCSK9 level is elevated in RA patients,which is related to RF,disease activity,TC,TG,LDL,VLDL,ApoB.This suggests that PCSK9 is potentially linked to inflammatory reaction and lipid metabolism in rheumatoid arthritis.

ABSTRACT

Objective To investigate changes in bone mineral density (BMD) and analyze its related factors in community populations to provide the early diagnosis of osteoporosis (OP) and give right guidance to prevent osteoporosis. Methods The quantitative ultrasound BMD analyzer was used to measure BMD of heel in 8 711 adults in community. At the same time a questionnaire survey was conducted among these subjects. The data were analyzed by Logistic stepwise regression analysis. Results With the age changing, males and females tended to have different BMD variation. Male and female's BMD peak values were respectively in 35-age group, 30-age group and decreased as age increased. T value of BMD was different between male and female (-1.40(-2.0--0.7)vs-1.3(-2.0--0.5))(P<0.001). Both men and women had a higher incidence of low bone mass (53.34%vs 47.46%), and the difference was significant (P<0.001), whereas the incidence of osteoporosis after 55 years of age between men and women was statistically significance (P<0.05). BMD was the lowest for female than for male in 50-age group, and the prevalence rate of OP was remarkably increased. Analysis of related factors of BMD showed that body mass index (BMI), age, diabetes and menopause were risk facts. Conclusion The BMD among adults is related to many factors, in which the age, BMI and menopause are the most important factors. Much attention should be paid to low bone mass phenomenon, timely monitoring, timely intervention, develop healthy working and living habits, is important to the prevention of osteoporosis and its complications.

ABSTRACT

Objective To study changes in serum homocysteine(Hcy) and its correlation with serum levels of folic acid and high sensitivity C-reactive protein(hs-CRP)in Tibetan patients with mildto-moderate Alzheimer's disease at various high altitude areas,so as to direct the clinical diagnosis and treatment of AD in plateau hypoxia environment Method 108 cases were divided into four groups:23 AD Tibetan patients at middle altitude(AD/middle altitude group)and 23 healthy Tibetan subjects (healthy/middle altitude group) in Qinghai-Tibet Plateau Xining region,altitude at 2,260 m,31 AD Tibetan patients (AD/high altitude group)and 31 healthy Tibetan elderly subjects (healthy/high altitude group)in Yushu region at altitude of 3,800 m.Among the total study subjects,half are males,aged from 60 to 85 years.The levels of serum Hcy,Vitamin B12 and folic acid(FA)were measured by the Fluorescence Polarization Immunoassay(FPIA).Serum hs-CRP,triglyceride(TG)and low density lipoprotein cholesterol(LDL-C)were measured by automatic biochemistry analyzer.Correlation of Hcy with FA and hs-CRP was analyzed.Result Both high altitude and middle altitude group showed the levels of Hcy and hs-CRP were significantly higher in AD Tibetan patients than in healthy control at the same altitude(allP<0.05).The levels of Hcy,LDL-C and hs-CRP of subjects were higherat high altitude than at middle altitude(P<0.05).In contrast,folic acid levels in AD and control groups were lower at the high altitude than at middle altitude(P<0.05).The levels of vitamin B12 and TG were not significantly different among all four groups.Multiple logistic regression analysis showed that altitude,folacin and hs-CRP were the risk factors for Hcy in patients with AD at plateau(OR =0.351,2.794,3.021,P=0.045,0.037,0.016).Conclusion Along with increased altitude,serum level of Hcy is significantly increased in AD Tibetan patients living in high altitude area.High altitude,high hs-CRP and lower folacin may be the risk factors for hyper-homocysteine in AD Tibetan patients with high altitude,and their combined effects are involved in the occurrence and development of AD.

Objective To observe the association between the polymorphism of PINK1 gene exon 4 T313M and Parkinson's disease ( PD) in Xinjiang Uygurs.Methods The genetic DNA was extracted from 175 Uygur patients with PD (PD group) and 163 Uygur healthy controls (control group).The PINK1 gene exon 4 T313M was amplified by PCR.The genotype and allele were determined by restriction enzymes enzyme technology, and then validated by sequencing analysis.Results The frequency of PINK1 gene T313M T/T genotype in PD group was 2.9%, which of C/C genotype was 97.1%;the frequency of PINK1 gene T313M T allele in PD group was 2.9%, which of C allele was 97.1%.The frequency of PINK1 gene T313M T/T genotype in control group was 1.2%, which of C/C genotype was 98.8%;the frequency of PINK1 gene T313M T allele in control group was 1.2%, which of C allele was 98.8%. The difference of the polymorphism of PINK1 gene T313M between the two groups had no statistical significance ( P>0.05).Conclusion The polymorphism of PINK1 gene exon 4 T313M has nothing to do with the genetic susceptibility of Xinjiang Uighur PD patients.

ABSTRACT

Objective To investigate the clinical efficacy of trimetazidine in treatment of ischemic cardiomy-opathy heart failure and its influence on cardiac function and heart rate variability .Methods 160 cases of patients with ischemic cardiomyopathy heart failure were randomly divided into the two groups with 80 cases in each group .The two groups were given conventional treatment of anti heart failure ,the observation group used trimetazidine on basis of that,clinical effect and the influence on the index of heart rate variability and cardiac function of two groups were ob -served.Results The total effective rate of the observation group was 91.25%,significantly higher than 73.75% of the control group (χ2 =9.57,P<0.05);LVEDD and LVESD of the observation group after treatment were (49.88 ± 4.60)mm and (37.10 ±3.15)mm,significantly reduced than before treatment (t=7.44,7.83,all P<0.05) and the control group (t=6.60,6.85,all P<0.05); while the LVEF was (48.04 ±4.20)%,increased significantly than before treatment (t=6.51,P<0.05) and the control group (t=7.11,P<0.05);SDNN,SDANN,rMSSD and PNN50 of the observation group after treatment were (124.72 ±7.02)ms,(111.20 ±5.89)ms,(46.74 ±4.22)ms and (17.72 ±2.08)%,increased significantly than before treatment (t=7.43,7.83,7.06,8.17,all P<0.05) and the control group (t=6.66,6.90,6.15,7.21,all P<0.05).Conclusion Trimetazidine has an exact effect in treat-ment of ischemic cardiomyopathy heart failure ,which can improve heart function and heart rate variability which has an important value in clinical application .

Objective To study the efficacy and safety of Fasudil in treatment of elderly diastolic heart fail -ure related pulmonary hypertension ,and provide evidence for clinical treatment .Methods 70 cases of elderly dias-tolic heart failure related pulmonary hypertension were divided into the study group and control group of 35 cases in each group according to random number table methods ,the two groups were given beta blockers ,angiotensin converting enzyme inhibitors and other drugs for treatment ,the study group was given fasudil on basis of that ,6min walking dis-tance,pulmonary artery systolic pressure ,arterial partial pressure of oxygen ( PaO2) of two groups before and after treatment as well as the changes of blood pressure and heart rate of the study group before and after treatment were compared.Results Before treatment,6min walking distance,pulmonary artery systolic pressure and PaO2 of the two groups were (290.3 ±1.3)m,(83.3 ±0.9)mmHg,(66.3 ±1.4)mmHg and (289.2 ±1.1)m,(82.9 ±0.8)mm-Hg,(65.9 ±1.3)mmHg showed no statistical significance (t=2.392,4.927,5.132,P=0.073,0.069,0.063),after treatment,6min walking distance ,pulmonary artery systolic pressure and PaO 2 of the study group were (410.4 ±2.3) m,(60.1 ±1.1) mmHg,(89.3 ±0.6) mmHg significantly better than those of the control group ,with statistically significant difference(t=8.918,9.142,11.319,P =0.024,0.019,0.017);the systolic blood pressure,diastolic pressure and heart rate of the study group before and after treatment were (132.8 ±2.1)mmHg,(84.3 ±1.1)mm-Hg,(69.2 ±1.4) times/min and (133.2 ±1.9) mmHg,(83.9 ±0.9) mmHg,(68.9 ±1.3) times/min showed no statistical significance(t=4.193,4.018,3.987,P=0.061,0.078,0.068).Conclusion Fasudil has a better effect in treatment of elderly diastolic heart failure related pulmonary hypertension ,which can significantly reduce the pulmo-nary artery pressure ,and has a little effect on the heart ,with high safety .

Objective To analyze the relationship between the urine iodine level in different pregnant periods of women and the thyroid function .Methods The pregnant women were randomly sampled in Foshan city and 490 cases were selected as the research subjects ,including 170 cases of early stage pregnancy ,162 cases of middle stage pregnancy and 158 cases of late stage pregnancy . According to the thyroid function ,490 subjects were divided into the normal thyroid function group and the abnormal thyroid func‐tion group .Results 140 cases(88 .61% ) in the late stage pregnancy had normal thyroid function ,which were less than 166 cases (97 .65% ) in the early stage pregnancy and 157 cases (96 .91% ) in the middle stage pregnancy ;the constituent ratio of urine iodine < 100 μg/L in the early stage pregnancy was 19 .88% (33/166) in the normal thyroid function ,which was lower than 75 .00%(3/5) in the abnormal thyroid function group .The constituent ratio of urine iodine 100 - 300 μg/L in the early stage pregnancy group was 56 .0% (93/166) ,which was higher than 0 .0% (0/4) in the of abnormal thyroid function group .Conclusion It is neces‐sary to conduct the urinary iodine monitoring in early pregnant woman ,moreover it is suggested that the thyroid function detection will be conducted in the pregnant women with urine iodine < 100 μg/L and urine iodine 100 - 300 μg/L .

ABSTRACT

Objective To analyze the clinical effect of atorvastatin combined with conventional cardiovascu-lar drugs therapy in treatment of coronary heart disease.Methods 200 patients with coronary heart disease were col-lected.They were randomly divided into the observation group(n =100)and control group(n =100).The observation group was given atorvastatin combined with cardiovascular routine treatment for 1 year,the control group was given cardiovascular routine therapy for 1 year.The clinical effect and serum levels of TC,HDL -C,LDL -C were ana-lyzed.Results After treatment,the total effective rate of the observation group was 95%,which was significantly higher than 75% of the control group (χ2 =16.33,P =0.00 0.05;HDL -C:(1.35 ±0.23)mmol/L vs (1.29 ±0.32)mmol/L, t =1.51,P =0.15 >0.05;LDL -C:(4.54 ±0.62)mmol/L vs (4.48 ±0.61)mmol/L,t =0.84,P =0.20 >0.05]. After treatment,the serum levels of TC,HDL -C,LDL -C of the observation group were significantly better than those of the control group[TC:(3.39 ±0.31)mmol/L vs (5.78 ±0.64)mmol/L,t =2.36,P =0.02 <0.05;HDL -C:(1.63 ±0.42)mmol/L vs (1.32 ±0.51)mmol/L,t =2.87,P =0.00 <0.05;LDL -C:(2.37 ±0.42)mmol/L vs (3.95 ±0.43)mmol/L,t =2.62,P =0.01 <0.05].Conclusion Atorvastatin can reduce blood fat,improve the prognosis of disease,it is superior to the traditional treatment.

ABSTRACT

Objective To observe the clinical efficacy of Oseltamivir combined with Chinese medicine for the treatment of influenza. Methods One hundred and ninety-seven cases confirmed as influenza were randomly divided into treatment group ( N=101) and control group ( N=96) . Both groups were given conventional symptomatic and supportive treatment. Additionally, the treatment group received Oseltamivir and Chinese medicine based on syndrome differentiation, and the control group was given Oseltamivir orally. The treatment lasted 5 days. The clinical outcomes included incidence of flu symptoms, time for body temperature decreasing to the normal and time for the relief of all symptoms in both groups. Results ( 1) The total effective rate of the treatment group was 98.0%, superior to 91.7%of the control group (P<0.05) . (2) The treatment group had an effect on sub-siding fever and relieving symptoms more quickly than the control group, the differences being significant (P<0.05 or P<0.01) . ( 3) The treatment group could relieve flu symp toms of sore throat, muscular soreness, cough, nasal obstruction and discharging more effectively, and the differences were significant compared with the control group (P<0.05) . Conclusion The overall effect of Oseltamivir combined with Chinese medicine is better than Oseltamivir alone, showing the advantages of subsiding fever in a relatively short period of time and relieving flu symptoms more effectively.

ABSTRACT

OBJECTIVE: To study the reaction yield of SC-mPEG with bovine hemoglobin(BHB) under different conditions so as to establish a method for the analysis of the SC-mPEG-BHB mixtures.METHODS: The product was analyzed with the methods of SDS-PAGE,GPC,CE,TNBS,etc.The optimal reaction system for SC-mPEG with BHB was established based on the effects of different factors on SC-mPEG-modified BHB.RESULTS: The optimum reaction conditions for SC-mPEG with BHB were established as follows: SC-mPEG and BHB were mixed at an optimal ratio of 60∶1 and the reaction was lasted for 2 h in(pH9 and 4 ℃) buffer solution.CONCLUSIONS: This study established a foundation for the following study of red blood cell substitutes.
